In A section We have added a Select Item dropdown and Checked Items read only text input filed.
On click dropdown it will launch a local action in overlay and inside the overlay configured a repeating dynamic layout with data page. The section included in the repeating dynamic layout hold 2 things,
1) a check box property to capture multiple selection
2) value of the selected item.
when ever user select a dropdown and select multiple check boxes. On each selection of checkbox selected value has to show in comma separated in the checked items read only field.

It's not clear what your question is.
But what you are describing is very similar to the Multi-Select control provided OOTB.

Move checked items field to a new section. Include this new section after selected items drop down box.
In the onChange event of checkbox (multi select section), use refresh section with above created section name. Create a new DT to be used in this action like below.
In the DT, prepare CSV of selected check box values
If you have a page list of all selected check boxes, use below function to get csv out of it
@pxDataManipulate.pxStringCSVFromPageList(pxThread.pyUserPortals,"pyLabel")
Otherwise, Loop through the page list property , check if property is selected and if so, add it to custom parameter like below. Set this parameter value to your checked items property.
